MK-677 (ibutamoren) is an orally active non-peptide ghrelin receptor agonist. It's technically a small molecule rather than a peptide but is grouped with peptide GH-secretagogues because of its mechanism. Under FDA review for similar regulatory considerations.
How it works

MK-677 binds the ghrelin receptor with sufficient duration and oral bioavailability to produce sustained GH/IGF-1 elevation from once-daily oral dosing. This distinguishes it operationally from injectable peptide GH-secretagogues.

Research history.
MK-677 was developed by Merck in the 1990s and reached Phase 3 trials for elderly frailty before development was discontinued. Pharmacokinetic and short-term efficacy data exist. Cannot currently be compounded under 503A.
